Natural Products – Methods and Optimal NMR Techniques
Introduction to Natural Products for Drug Discovery Research and Beyond
Topics:
- Sourcing of raw material
- Extraction techniques
- Multi-step purification techniques for screening and NMR data acquisition
- Sample preparation and handling at the microgram scale
- NMR Probe options
- NMR sample preparation and handling at the micro-liter scale
- NMR experiments and optimizations
- Data manipulation and interpretation
Remarks:
From isolation, purification to structure elucidation, natural products presents many different and varied challenges in creating pure samples from limited biomass. The course offers an introduction into the complexities of working with natural products from a drug discovery perspective. This course will focus mainly on plant biomass but can be extrapolated to include any natural product source. The user will experience all of the analytical techniques included in a patented (US Patent 7,367,933) natural product library production process, to isolate NMR ready samples. From here, the course will cover working with compounds at the microgram and micro liter scale, from sample creation to sample transfer. Basic and optimal NMR experiments will be covered including post processing of all NMR data. Structure elucidation approaches will include starting points of software choices, data manipulation and structure proposal and verification.
